The Royal pair created the official page under the handle SussexRoyal on the social media platform yesterday.
They reached one million followers just six hours later by 9pm last night.
The previous record was held by Korean pop star Kang Daniel at 11 hours and 36 minutes.
Meghan, 37, and Harry, 34, unveiled a set of new pictures in their first post with a Royal monogram.
They wrote: “Welcome to our official Instagram; we look forward to sharing the work that drives us, the causes we support, important announcements, and the opportunity to shine a light on key issues.
“We thank you for your support, and welcome you to @sussexroyal.”
The gallery includes nine of their favourite images, as well as a previously unseen photo of the Royal couple in Fiji during their tour last year.
They previously used the Kensington Palace Instagram account, joint with the Duke and Duchess of Cambridge.

It comes just two weeks after the Palace announced Meghan and Harry were separating from the Cambridges and getting their own royal household.
It “will ensure that permanent support arrangements for the Duke and Duchess’s work are in place as they start their family and move to their official residence at Frogmore Cottage”.
Meghan and Harry are also expecting their first child in the next few weeks.
Irish bookies Paddy Power believe we still have a little while to wait before the baby arrives – with between April 22-29 the current favourite at 2/1.
There is also a chance that the new arrival could share a birthday with their great-grandma, the Queen, if the birth is on April 21st (7/1).
